Promotes sleep
Melatonin is best-known for its role in sleep regulation. It signals to the body that it is time to prepare for nocturnal rest by creating the feelings of drowsiness and sleepiness. Melatonin also lowers body temperature to encourage sleep

Melatonin is crucial to synchronise the internal biological clock which controls physiological cycles. It allows the body to adjust to natural variations in light throughout the day, waking us in the morning and preparing us for sleep in the evening. Further, almost all bodily functions (metabolism, immune system, blood pressure, etc.) are subject to the circadian rhythm, which explains the importance of this molecule for the proper functioning of the body. In human beings, the central clock is found in the suprachiasmatic nuclei of the hypothalamus

L-Tryptophan is an essential amino acid, a building block of protein and necessary for various biological functions. Its chemical structure consists of an indole nucleus – a six-membered aromatic ring, attached to a side chain of amino acids.
It is said to be essential as the human body cannot synthesise L-Tryptophan itself, therefore it must be supplied through diet.
Tryptophan is essential to melatonin synthesis as it forms the first chain in a series of biochemical reactions leading to the production of this sleep hormone. Working in synergy, tryptophan and melatonin help regulate sleep and the biological clock
Vitamin B9
Vitamin B9, also known as folic acid, is a water-soluble vitamin essential to various biological functions
The methylated form of vitamin B9, known as 5-methyltetrahydrofolate (5-MTHF), is important because it represents the biologically active form. It consists of a pteridine ring, a p-aminobenzoic (PABA) group, a glutamic acid residue and a methyl group (-CH3)
Vitamin B9 is essential to the melatonin metabolism. It is involved in the hydroxylation of tryptophan, producing 5-HTP, then during the methylation of serotonin, producing melatonin. It therefore plays a role in the first and final steps of the metabolic pathway of melatonin synthesis
Vitamin B6
Vitamin B6 is a water-soluble vitamin present in three forms: pyridoxine, pyridoxal and pyridoxamine
Its active form in the body and the form used in biological reactions is pyridoxal 5’-phosphate or PLP
Its chemical structure consists of a pyridine ring with a pyridoxal group in position 4, a phosphate group in position 5, an aldehyde group in position 1 and an amino group (-NH2) in position 2.
The synergy between vitamin B6 and melatonin lies mainly in the fact that vitamin B6 is an essential co-factor in the synthesis of melatonin for the conversion of 5-hydroxytryptophan (5-HTP) into serotonin, then serotonin into melatonin

Vitamin B12, also known as cobalamin, is an essential vitamin with a very complex structure. This vitamin contains a colbalt (Co) ion at the centre of a corrinoid ring
The methylated form of vitamin B12 is known as methylcobalamin. Its structure is similar to that of vitamin B12 cobalamin with a methyl group (-CH3) attached to the cobalt ion. This form is said to be bioactive and corresponds to the active and metabolically functional form of vitamin B12 which is present in the body and plays a role in various metabolic processes
