Vitamin B6
A water-soluble vitamin present in three forms: pyridoxine, pyridoxal, and pyridoxamine. Its active form in the body is pyridoxal-5-phosphate or PLP. Vitamin B6 is an essential cofactor in the synthesis of melatonin, enabling the conversion of 5-hydroxytryptophan (5HTP) into serotonin and subsequently serotonin into melatonin.
L-Glycine
L-Glycine is an amino acid whose therapeutic potential in sleep has been increasingly studied in recent years. Research has shown that taking glycine before bedtime reduces sleep onset time and improves sleep quality, particularly during the REM phase. Glycine seems to influence central body temperature, promoting hypothermia necessary for sleep onset. This mechanism is linked to its action on NMDA receptors in the brain’s suprachiasmatic nucleus, the primary regulator of circadian rhythms.
Vitamin B9
A water-soluble vitamin also known as folic acid. The methylated form of Vitamin B9, called 5-methyltetrahydrofolate (5-MTHF), represents the biologically active form. Vitamin B9 is involved in melatonin metabolism during the hydroxylation of tryptophan into 5-HTP and the methylation of serotonin into melatonin.
Melatonin
Melatonin is a hormone naturally produced in the pineal gland of the brain. Chemically, it has a simple structure composed of an indole ring and a methoxy side chain. Melatonin is closely associated with regulating circadian rhythms and plays a central role in the sleep-wake cycle. Its production is influenced by light and darkness, making it a key hormone for internal biological clock synchronization.
